All right... So I gave this a try with Manchu last night. I used the Assimilated Engine, the Obelisk Warp Core, and the Polaric Inverter Eng Console. Anything that will increase base warp speed, and reduce cooldown and/or increase turn rate in Slipstream seems to be the way to go. Transwarp is pretty irrelevant. Here's the path I took. I started in the lower left of the Beta Quadrant, and followed this line, using the astrometrics display so I could see the borders of the sectors as I was flying. Blue is slipstream, and red is warp. This is in my Odyssey, by the way, which will slipstream for twice as long as most ships, but I believe there are a few other ships with this same ability. Once I finished with the Beta Quadrant, I used the Transwarp to Defera to keep going.

So turns out now you've got to fly past a series of specific star systems... that complicated things a bit. I did figure out a path that does work. I completed Tour the Galaxy with about 30 seconds left on the clock.
Same gear as before. Assimilated Impulse (increases warp to over 10, which is necessary), Obelisk Warp Core (reduces cooldown on SS and increases turn rate in SS), and the Polaric Inverter from Delta Flight. Added the Leadfoot trait (can be found on the exchange for around 100,000EC), and was generously granted Diplomatic Immunity and Raiding Party. I have no Driver Coil inate skill, but my warp speed was topping out at warp 21. I suspect that having Driver Coil maxed out could push this upward clear of warp 30, possibly negating the need for slipstream at all. My slipstream lasted twice as long as normal with the Odyssey cruiser. A similar ship would be beneficial, if you've got it. I know the Vesta has the same ability. Not sure about others.
I think that's about it for relevant gear.
I started out to the west of the Breen system in the Alpha Quadrant, activated the mission, and used slipstream to get to Ferenginar. When approaching a system be sure to "wiggle" the ship to the left and right so the autopilot won't automatically stop short of that system.
Once I entered the Ferenginar system, I changed instances to the emptiest one I could quickly find in the Alpha Quadrant, which repositions the ship instantly to near DS9.
Manually flew to DS9, then full stop, and turned toward Algira. From here, activate SS whenever available and proceed Algira, Arawath, Defera, Tzenketh, Mizar, Corwin, Betazed, Chapel, Tellar, then angle east to enter the Beta Quadrant as soon as possible (should be just east of Tellar)
At this point, I had about 10 minutes left.
You enter the Beta Quadrant near Teneebia (should be just to the northeast). Continue in a wide spiraling arc through Teneebia, Nimbus, Lirss, Alini, Vorn, Iconia, Pheben, Gamma Eridon, Mylasa, New Romulus, Virinat, Argelius, and Vendor.
This should clear all of Romulan space, leaving you in the center of the quadrant. Now, instance change again to the most empty instance (I think this may help loading time)
You should then be near Sol, pointed south, with about 4 minutes left on the clock.
SS or maximum warp south toward Orion, Risa, Traelus, Drozana, Archanis, Khitomer, Qo'NoS, and Sanek.
